ABSTRACT

Objective:Through the establishment and preliminary application of standardized, real-time and online database of severe sepsis in children, we can realize the information about management of children with severe sepsis, providing data to support scientific research and clinical work, and building a homogeneous scientific research and clinical platform for multiple units participating in constructing the database.Methods:We designed, developed and established an online database system for children with severe sepsis, which was filled in in real time by 19 member units of Diagnosis and Treatment Collaboration Group for Sepsis in Children in Shandong Province.The basic information, treatment and prognosis of children entered in the database were preliminarily analyzed.Results:(1)A real-time online disease-specific database for severe sepsis in children was established primarily, with the classification, storage, logical retrieval, statistical analysis and map-making of the information of children with severe sepsis.(2)Further analysis was conducted on the clinical data of 602 children with severe sepsis, which had been recorded in the database.There were 341 males(56.6%)and 261 females(43.4%). The median age was 16 months.There were 230(38.2%)children younger than 1 year old, and 274 children aged from 1 to 5 years old(45.5%), 98 children(16.3%)were 6 years old and above.Three hundred and forty-three cases(57.0%)were from rural areas.One hundred and eighty-one cases(30.1%)were found to have basic diseases.The primary site of infection was the respiratory system(40.8%). The in-hospital mortality was 15.6% in children with severe sepsis.Multivariate Logistic regression analysis showed that high pediatric risk of mortality score Ⅲ score at 24 hours after admission, acute respiratory distress syndrome, acute renal injury, mechanical ventilation and vasoactive drug treatment were independent risk factors for in-hospital death in children with severe sepsis( P<0.05). Conclusion:The disease-specific database of severe sepsis in children can provide data for scientific research and clinical practice of sepsis management in children, and contribute to the formulation and improvement of clinical decision-making in the future.Simultaneously, it has also constructed a homogeneous scientific research and clinical work platform, which may contribute to the common development of the participatory units.

ABSTRACT

The concentrations and distributions of trace metals and rare earth elements (REE) in sediment and mussel samples collected from the India Ocean hydrothermal area were analyzed.The metal correlation between organisms and sediments was investigated, and the ecological and chemical characteristics of REE were also explored.The results showed that, the trace metals in sediments were mainly Fe (96.6 mg/kg), Mn (1.14 mg/kg) and Zn (322.6 μg/kg), and Fe had high ratio of 98.15% by normalized calculation, which indicated that the available sediments in this studying hydrothermal area mainly consisted of iron ore substances.Trace metals and REE distributions all had good correlation between deep-sea sediments and deep-sea mussels, and the correlation coefficients were 0.991 for trace metals and 0.996 for REE.The contents and distributions of metal elements in deep-sea mussels were different from those in offshore mussels.The REE distributions in sediments and mussels showed obvious fractionation phenomenon, and the enrichment of LREE in mussels was significant.Through the REE patterns, Eu and Gd in sediments and mussels all showed anomalies, and Eu had a significant abnormal phenomenon in deep-sea sediments and deep-sea mussels.Besides, δEu values were 9.50, 10.68 and 0.23 in deep-sea sediments, deep-sea mussels and offshore mussels, respectively, and δCe were 2.21, 2.71 and 4.38, which showed that the enrichment sources of REE in offshore mussels and deep-sea mussels were different, and the REE in sediments and mussels from the India Ocean were homologous.

ABSTRACT

Objective To investigate the expression of P16 and CD44 in gastrointestinal stromal tumors (GIST) and their relationship with the prognosis of patients. Methods The GIST specimens of seventy patients who received surgical excision were collected. Tissue microarray of the seventy GIST samples was constructed. The expression of P16 and CD44 were detected by the immunohistochemical staining. The patients were followed up via out-patient examination and telephone. Results All the patients were followed up for 2-212 months, and the median time for follow-up was 68 months. The differences of the expression of P16 in GISTs among NIH risk ranks were insignificant (P > 0.05). The differences of the expression of CD44 in GISTs among NIH risk ranks were statistically significant (P < 0.05). Univariate analysis showed that tumor size, mitotic count, tumor location, NIH risk rank, the expression of P16 and CD44 were related to the prognosis of GIST patients. Multivariate showed that tumor size, mitotic count, tumor location, and the expression of CD44 was independent prognosis factors of GIST patients. Conclusion CD44 could be used as a biomarker in predicting the prognosis of GIST patients.

ABSTRACT

The particles suspended in seawater have great influence on pollutant migration and transformation in marine environment, while the lipophilic algae toxins enriched by the particles suspended in seawater will lead more serious toxicity to marine filter feeders. In this study, a new method was developed for the simultaneous determination of eight lipophilic algae toxins in suspended particles by ultra performance liquid chromatography-tandem mass spectrometry ( UPLC-MS/MS ) . After extracted with methanol by ultrasonic-assisted extraction, the sample was separated on an Acquity UPLC BEH C18 column (50 mm×2. 1 mm, 1. 7 μm) using gradient elution of acetonitrile and water containing 5 mmol/L ammonium acetate as eluent modifiers. The qualitative and quantitative analyses were carried out by electrospray ionization ( ESI) tandem mass spectrometry in multiple reaction monitoring ( MRM) mode. Under the optimal conditions, satisfactory precision (relative standard deviations (RSD≤14. 1%), recoveries (83. 8%-110. 4%) and detection limits (2. 9-103 pg/g) of the method were achieved. Good linearity (R2≥0. 99) was also obtained for all studied analytes. Then, the method was applied to determine the amounts of the eight lipophilic marine toxins in authentic suspended particle samples collected from Qingdao near-shore area. Pectenotoxin 2 ( PTX2 ) was detected in the samples from Shilaoren beach and No. 3 bathing beach with concentration ranges of 717 and 790 pg/g, respectively.

ABSTRACT

A microwave digestion system was preparation for the digestion of the sediment samples of 16 stations in the Southe mid_Atlantic ridge by using HNO3_H2 O2_HF as the digestion reagent. The rare earth elements ( RE ) in sediments were determined by inductively coupled plasma mass spectrometry, and distribution characteristics of rare earth elements were studied. The microwave digestion_ICP_MS method was used for the determination of rare earth elements with a good linear relationship ( r=0 . 9997-1 . 0000 ) for each element. The detection limit reached ng/L level, the relative standard deviation ( RSD, n=3 ) was less than 3% and the relative error was 6%. The total amount of rare earth elements (ΣRE) in sediment samples from 16 stations varied in the range of 37. 25-134. 77 μg/g, the ratio range of light RE/heavy RE ( LRE/HRE) was 0 . 61-1 . 70 , the average value was 1 . 27 , and the enrichment of light rare earth elements in sediments was slightly obvious. The RE distribution patterns were basically the same in each station with obvious fractionation between LRE and HRE. The RE distribution patterns were also similar in sediments from different sources with slightly difference between terrestrial and marine sediments. The δEu and δCe in the sediments had negative anomaly which showed that the rare earth elements in sediments came from the seawater. This study first analyzed the content and distribution of rare earth elements in the southern Atlantic, providing data and technical support for further study of the distribution of rare earth elements in the Atlantic.

ABSTRACT

SuperparamagneticSiO2@Fe3O4microspherewithcore-shellstructurewaspreparedbythe method of hydro-thermal and St?ber method, and modified by (3-aminopropyl) triethoxysilane ( APTES) . The obtained amino-Fe3 O4@SiO2 microsphere with controlled surface charge properties was characterized by SEM and TEM, and the surface electric property of amino-microsphere was investigated by Zeta potential measurement. The microsphere was used to extract DNA from human blood, and the solid phase extraction method by microspheres was developed. The mechanism of action between Aminated-Fe3 O4@SiO2 microsphere and DNA was explored, and the gel electrophoresis and PCR test were done on the extraction product. The result showed that genomic DNA with high purity was successfully extracted from whole blood by using amino-Fe3 O4@SiO2 microsphere, with the extraction rate about 70%, the saturated adsorption capacity for DNA was about 40 ng/μg, and the elution could be directly used for further bio-analysis.

Around the core issue how to evaluate and test hypothesis, starting from the concept of hypothesis and scientific hypothesis, learning form recent progress and understanding at home and abroad, we have analyzed the type of scientific hypothesis, test methods and pre-mortem and post-mortem evaluation criteria in depth, established scientific vision and discipline ruler, explored the awareness level of the Chinese medicine hypothesis, pointed out the problems and gaps. Whereby, we have analyzed and demonstrated brain regulating five internal organs concept of wholism scientific hypothesis, and proven it belongs to the scientific hypothesis. We were confident that these works mentioned above would lead the theoretical reform changing Chinese medicine hypothesis from traditional description to the nature clarification.

ABSTRACT

To guarantee the research and establishment of the modern basic theory of traditional Chinese medicine, We firstly proposed the concept of the academic standards of the modern basic theory of traditional Chinese medicine and its logical structure and standards points in this paper. According to the research progress at home and abroad, we explored the basic concept of academic, academic standards, academic of Chinese medicine, academic standards of Chinese medicine in depth, given the logical definition of relevant concepts, and revealed the connotation and logical hierarchy relationship between different concepts. On this basis, we proposed three-level standards points of the academic standards of Chinese medicine. All of these works mentioned above would provide a preliminary standards protection for the academic research of traditional Chinese medicine and the establishment of the modern basic theory of traditional Chinese medicine.

ABSTRACT

Objective To investigate the features and diagnostic value of computed tomography ( CT) in cys-titis glandularis .Methods The CT images and clinical data of 9 patients with cystitis glandularis confirmed by histol-ogy were studied retrospectively .Results The characteristics of cystitis glandularis ( location,shape,density and the relationship with adjacent tissues)were clearly displayed on CT.The lesion was located in the bladder trigone(n=5), right anterior wall(n=1),left posterior wall(n=2),diffuse lesion(n=1).According to the shape,8 lesions showed mass type,the range of these lesions were limited and some lesions were combined with cystic degeneration ,only one lesion showed a diffusely thickened bladder wall .After enhancement ,CT value increased from 14.0-45.0 HU to 25.0-62.0 HU.Conclusion CT has high sensitivity and accurate in diagnosis of cystitis glandularis .

ABSTRACT

Based on the selective permeability of cationic exchange membrane and the characteristic of electri cal driven, a new and non-damage apparatus that can be used to separate compounds with different electric property was built to capture and enrich DNA.λ-DNA, as a model compound, was used to study the charac teristic of capturing and enriching DNA in this device.Factors that might affect the efficiency such as voltage and flow speed were optimized and gel electrophoresis was done to study if DNA was denaturation, PCR was done to prove the concentration can be used to further bioanalysis.Results were got; the apparatus could used to capture and enrich DNA scathelessly under the condition of low-voltage, the concentrate multiple was 62, and the recovery efficiency was accessible to 47%, concentration can be used for the next bioanalysis.Auto matic, easy to handle and little organic reagent using made it be advantageous, it could be miniatured to com bine with other biochips.This method had a large application space in field of DNA extraction, purification and concentration.

ABSTRACT

A new method based on high performance liquid chromatography-electrospray ionization time of flight-mass spectrometry (HPLC-ESI-TOF/MS) was developed for the rapid identification of active compounds in Styela clava and the development of its specific chromatograms. Samples were extracted by ultrasonic-assisted extraction, and the extraction conditions were optimized. The developed HPLC-ESI-TOF/MS method was used to identify the components in Styela clava extract, and a specific chromatogram based on HPLC analysis was established. Ten compounds in Styela clava extract have been primary identified by HPLC-ESI-TOF/MS on-line detection combined with literature review. The result of similarity evaluation for specific chromatograms indicated that the quality of different Styela clava samples was not entirely consistent. This method has the advantages of simple operation, rapid measurement and it is a powerful tool for identification of active components in Styela clava and its quality control.

ABSTRACT

Mammal metallothionein(MT) folds into two separate domains that exhibit different structure and metal binding propertity independently, the study of the strategy of metal ions binding with MT would give better understanding of their exact biological functional mechanisms. In this study, a method using eletrospray ionization mass spectrometry (ESI-MS) phase liquid chromatography and identified by ESI-MS. Different amounts of Cd or Cu were then added in MT-2a samples and ESI-MS was employed to determine the mass difference of MT in different samples. The results Cd2+4S11; while Cd is attached in separate binding sites for the formation of Cd2+3S9 cluster, which intermediately formed with five and six Cd ions were detected. For the Cuprous ions, it prefers to cooperatively bind in β-domain with the form of Cu4-MTβ. The binding form in β-domain would convert from Cu4 into more Cu binding form with the addition of Cu. When high concentration of Cu was added in samples, the result suggested that

ABSTRACT

Arsenic and its species in dry seafood products(DSPs) Kelp, Sargassum fusiform, Laver, Enteromorpha prolifera, were systematically determined by high performance liquid chromatography-inductively coupled plasma mass spectrometry(HPLC-ICP-MS). It was found that the of arsenic species in botanic DSPs are three unknown arsenicals(U_2, U_3 and U_4), which are speculated as arsenosugars and take approximate 63.5% 92.7% of all arsenic species detected. These arsenosugars were indentified as dimethylarsinic(DMA)-glycerol ribose, DMA-phosphate ribose and DMA-sulfate ribose by their accuracy molecular weight 329.0599, 483.0738 and 409.0162, using direct confirmatory of HPLC-TOF-MS. The quantitative analysis of these arsenic species shows that although the total arsenic exceeds the National Standard value, most of the marine plants DSPs are safe to eat. The main component in most marine plants is DMA-sulfate ribose(51.1%-80.3%), while the main component of Kelp is DMA-phosphate ribose(48.9%). The sum of those three arsenosugars was 63.5%-92.7% of peak areas in all arsenic species detected.

ABSTRACT

The basical structure of diketopiperazines is a cyclic dipeptide condensed by two amino acids. Because of the stable framework of the six-member ring structure, and having two hydrogen bond donor and two hydrogen bond receptor, DKPs have become important chemical pharmacophores, with strong biological activities and pharmacological activities in the drug. A series of cyclic compounds were found from marine organisms in recent years, research showed that their functions are not limited on anti-bacterial, cytotoxic activity, and so on, but also playing an important role in regulatory mechanism of quorum sensing as signal molecules, they have become research hot point in ecological chemistry. This paper reviewed the research progress of diketopiperazines found in the marine microbial metabolites, and the future study trends was discussed and outlooked.

ABSTRACT

An online enrichment-separation system of C18 and inductively coupled plasma-mass spectrometry (ICP/MS) with flow injection (C18-FI-ICP/MS) was established for the determination of trace heavy metals in seawater. The optimized experimental parameters including pH,C18 column,concentration of methol in elute and the velocity of elute were investigated. This system was applied to the online preconcentration , separation and determination of trace heave metals in seawater samples successfully. The detection limit (3σ) for Cd, Pb, Co, Ni and Zn was 0.03, 0.09, 0.1, 0.1, 0.3 μg/L respectively. The RSD of Cd, Pb, Co, Ni, and Zn for six measurements of seawater was 6.8%,3.4%,1.3%,2.6%,0.5%,respectively. The accuracy was verified with spiked recovery test. The recovery for five elements was 91.3%,95.1%,100.4%,100.3%,95.2%, respectively.

ABSTRACT

Object A novel microwave-heated extraction (MHE) method was studied for the extraction of glycyrrhizic acid from Glycyrrhiza uralensis Fisch. Methods Several factors, such as temperature, time and microwave power were investigated and the appropriate MHE conditions were obtained from the orthogonal test. Under the optimum conditions, the optimal solvent was selected and the MHE was compared with ultrasonic extraction, leaching at room temperature and Soxhlet extraction,. Results The optimum conditions of MHE is extracting for another 40 min in 0.5% ammonia water after heated to 60 ℃ by microwave of 2 000 W. Yield of glycyrrhizic acid was about equal to that of Soxhlet extraction for 4 h, and that of leaching at room temperature for 44.3 h. Conclusion The MHE method is fast, efficient, energy-saving and high-selective, which is recommendable to the application to active compounds extraction from Chinese herbal medicines.
